بررسی فعالیت آنتیاکسیدانی و تعیین محتوای فنولی و فلاونوئیدی گیاه دارویی Tussilago farfara L. در طبقات ارتفاعی مختلف و مراحل فنولوژی |

مقدمه: گیاه علفی چندسالۀ پای خر (.Tussilago farfara L) از خانوادۀ Asteraceae است که برگها و گلهای آن خاصیت دارویی دارد و از مواد مؤثر آن برای معالجۀ سرفههای خشک و درمان التهابهای دهان، گلو و مجاری تنفسی استفاده میشود؛ بنابراین، هدف از این مطالعه بررسی برخی ویژگی های فیتوشیمیایی گیاه پای خر (Tussilago farfara L.) و مقایسۀ تغییرات آن در طبقات مختلف ارتفاعی و مراحل فنولوژی است. مواد و روش ها: برای بررسی روند تغییرات میزان مادۀ مؤثر گیاه موردبررسی در گرادیان ارتفاعی، با توجه به حضور و فراوانی گونه، طبقات ارتفاعی 1100 تا 2600 متری در استان گلستان انتخاب شد. بهمنظور نمونهبرداری از گیاه در مراحل فنولوژی (پیش از گلدهی، زمان گلدهی و پس از گلدهی) در هر طبقۀ ارتفاعی، بهطور تصادفی از گیاه در هریک از پنج طبقۀ ارتفاعی، نمونهها جمعآوری و به آزمایشگاه منتقل گردید. در پژوهش حاضر محتوای فنول کل با استفاده از روش فولین سیوکالتو، محتوای فلاونوئید با روش آلومینیوم کلرید و همچنین بررسی فعالیت آنتیاکسیدانی عصارۀ متانولی تهیهشده با استفاده از معرف DPPH صورت گرفت. یافتهها: نتایج نشان داد، بیشترین مقادیر محتوای فنول کل، فلاونوئید و فعالیت آنتیاکسیدانی در ارتفاعات بالاتر از 2200 متر است و اختلاف معنیداری میان طبقات ارتفاعی به لحاظ ویژگیهای فیتوشیمیایی وجود داشت. یافتههای تحقیق در بررسی و مقایسۀ خصوصیات فیتوشیمیایی گیاه در میان مراحل مختلف فنولوژی نشان داد، بالاترین مقدار محتوای فنول کل و فلاونوئید در مرحلۀ فنولوژی گلدهی و بالاترین میزان فعالیت آنتیاکسیدانی برگ گیاه در مرحلۀ رویشی با مقدار 08/42 درصد مشاهده شد. بحث و نتیجهگیری: نتایج این مطالعه اطلاعات ارزشمندی در تعیین دامنۀ ارتفاعی و مرحلۀ فنولوژی مناسب در برداشت برگ این گیاه، بهمنظور دستیابی به بالاترین مقادیر محتوای فنول، فلاونوئید و فعالیت آنتیاکسیدانی، برای دسترسی فعالان در زمینۀ گیاهان را دارویی فراهم آورد. |

Evaluation of Antioxidant Activity, Phenolic, and Flavonoid Content of Tussilago farfara L. in Different Altitudes and Phenological Stages |

Introduction: Tussilago farfara L. is a perennial forb belonging to the Asteraceae family. The leaves and flowers of this plant have medicinal properties, and its essential oil has been used for the treatment of dry coughs and inflammation of the mouth, throat, and respiratory tract. This study aimed to assess some phytochemical characteristics of Tussilago farfara L. and compare changes in this plant at different altitudes. Material & Methods: To investigate the changes in the amount of the effective substance of the studied plant in the altitude gradient, altitude classes of 1100-2600 meters were considered in Golestan province, Iran, considering the presence and abundance of the species. In each phenological stage (before and after flowering time), plant samples were randomly collected in each altitude class and transferred to the laboratory. Total phenol content and flavonoid content were measured using the Folin-Ciocalteu and aluminum chloride methods, respectively. The antioxidant activity of the methanolic extract was also evaluated using the DPPH (2,2-Diphenyl-1-picrylhydrazyl) reagent. Findings: The results showed that the highest amounts of total phenol content, flavonoids, and antioxidant activity were observed at altitudes above 2200 meters, and there was a significant difference in plants (Tussilago farfara L.) grown in different altitudes in terms of phytochemical properties. Based on the comparison of phytochemical properties of this plant in different phenological stages, the highest content of total phenol and flavonoids were observed in the phonological stage of flowering and the highest (42.08%) antioxidant activity of leaves was observed in the vegetative stage. Discussion & Conclusion: The results of this study can help activists in the field of medicinal plants to determine the appropriate height range and phenological stage in harvesting the leaves of this plant to achieve the highest levels of phenol content, flavonoids, and antioxidant activity. |
